How can I make:
http://mysite.com/Your_Account/profile=6.html

Be this instead:
http://mysite.com/{username}

Example, my current profile page on DFCMS:
http://dragonflycms.org/Your_Account/profile=HTML.html

What I want it to be:
http://dragonflycms.org/HTML

Server specs (Server OS / Apache / MySQL / PHP / DragonflyCMS):
Windows 8 / Apache: 2.2.24 / MySQL: 5.1.71 / PHP: 5.4.17 / Dragonfly: LATEST VERSION

example.com/string is already taken by modules.

You could try with string.example.com but few days ago I started looking at modifying the code to use subdomains for languages eg: en.example.com for Win and Linux OSs. Anyhow, this is not definitive and it may not make it so it still remains a valid choice.

Would you be ok with something like: example.com/u/HTML ?
This way it would be compatible with v10 where already written routers will also use shortcuts. You could then add a shortcut like: u -> name=Your_Account&profile=% to maintain compatibility.

CloudLinux / Apache 2.4 LSAPI / MySQLi 5.6 / PHP 5.6 / DCVS

This is the approach that I took: Can I further simplify a URL A ~ seems acceptably to me, and gives you something unique to search for.
